| Details | ACE-mediated deprotonation of cyanoacetyl-Phe-Phe leads to the formation of a ketenimine intermediate. This reactive intermediate, in turn, traps an active-site nucleophile to irreversibly inactivate the enzyme. This principle therefore seems to be suitable for the development of irreversible inactivators of therapeutically important zinc metalloproteases. |
